INTRODUCTION
The Ames assay assesses the mutagenic potential of test agents. The assay uses auxotrophic mutant strains Salmonella typhimurium or Escherichia coli. The S. typimurium strains ─ TA97, TA98, TA100, TA102, TA1535, TA1538 ─ have His- auxotrophic phenotypes due to mutations in histidine biosynthesis genes. The E. coli strains ─ WP2 uvrA or WP2 uvrA pKM101 ─ have Trp- phenotypes due to mutations in the tryptophan synthetic genes. Compounds are tested for activity to induce reversion of mutations to cause prototrophy (His+ or Trp+) which is detected as growth on minimal medium.
Procedure:
Test compound (3, 30, 300 and 3000 mg/plate) or vehicle are combined with bacteria, incubated with or without rat liver microsome enzyme homogenate (S9), then are mixed with molten agar (top agar) and poured onto the surface of a minimal glucose agar plate (bottom agar). The assays are performed in triplicate. The plates are incubated at 370C for 72 hours. Those cells which are prototrophic revertants grow and form visible colonies on the minimal medium.
An increase of compound-induced revertant colonies greater than 3 times that of spontaneous (vehicle-treated) revertant colonies indicates significant mutagenic activity. Colonies numbering ≤50% of the vehicle control indicates cytotoxicity.
