一、 背景与来源说明 / Background & Origin
NPC43 是一种来源于人鼻咽癌(Nasopharyngeal Carcinoma, NPC)患者原发肿瘤组织的永生化肿瘤细胞株。由于大多数常见鼻咽癌细胞株在体外长期传代过程中容易丢失 EB 病毒(Epstein-Barr Virus, EBV),NPC43 是少数能够稳定携带并维持 EB 病毒(EBV-positive)的患者来源鼻咽癌细胞模型之一。NPC43 细胞株广泛应用于 EBV 阳性鼻咽癌发病机制、EB 病毒潜伏感染与致癌基因表达、肿瘤免疫微环境、放化疗敏感性及靶向药物筛选的研究。

三、 培养条件与传代指南 / Culture & Handling Protocols
1. 完全培养基配方 / Medium Formulation
基础培养基 / Basal Medium:RPMI-1640 基础培养基
血清与添加剂 / Serum & Supplements:
10% 至 15% 胎牛血清 (FBS)
ROCK 抑制剂(如 Y-27632,4 至 10 uM,依建系与消化复苏需要添加)
抗生素 / Antibiotics:100 U/mL 青霉素 + 100 ug/mL 链霉素
2. 培养与传代操作 / Incubation & Subculture
培养环境 / Conditions:37 摄氏度,5% CO2,饱和湿度
密度管理 / Confluence:保持细胞密度在 30% 到 85% 之间,避免过度汇合导致 EB 病毒脱落或细胞自发凋亡
传代步骤 / Procedure:
吸除旧培养基,用 PBS 洗涤一次。
加入 0.25% Trypsin-EDTA,在 37 摄氏度下孵育 2 到 4 分钟至细胞脱落。
加入含血清完全培养基终止消化并轻柔吹打均匀。
以 200 x g 离心 5 分钟,重悬后按 1:2 至 1:3 比例接种。

BioVector® NPC43 Human Nasopharyngeal Carcinoma Cell Line
1. Background and Origin
NPC43 is a patient-derived cell line established from human nasopharyngeal carcinoma (NPC) tissue. Unlike many classical NPC lines that tend to lose the Epstein-Barr virus (EBV) during prolonged in vitro passage, NPC43 is one of the rare cell models that stably retains the EBV genome (EBV-positive). NPC43 serves as a vital model for investigating EBV-driven nasopharyngeal oncogenesis, viral latency/reactivation, tumor immunology, and therapeutic drug screening.

3. Culture and Handling Protocols
Culture Medium Formulation
Basal Medium: RPMI-1640
Serum and Additives: 10 to 15 percent Fetal Bovine Serum (FBS), supplemented with ROCK inhibitor (Y-27632, 4 to 10 uM) as required for lineage maintenance
Antibiotics: 100 U/mL Penicillin and 100 ug/mL Streptomycin
Incubation and Subculture Guide
Incubation Conditions: 37 degrees Celsius, 5 percent CO2, humidified atmosphere
Confluence Management: Maintain cell density between 30 percent and 85 percent to prevent EBV loss or cell detachment
Subculture Procedure:
Remove spent media and wash once with PBS.
Add 0.25 percent Trypsin-EDTA and incubate at 37 degrees Celsius for 2 to 4 minutes until cells detach.
Neutralize with complete medium and mix gently.
Centrifuge at 200 x g for 5 minutes, resuspend, and seed at a ratio between 1:2 and 1:3.
