BON-1(或 BON)是一种来源于人胰腺神经内分泌肿瘤(Pancreatic Neuroendocrine Tumor, pNET)的永生化细胞株。该细胞系保留了功能性神经内分泌细胞的关键生化特征,能够自发表达并分泌多种生物活性肽和神经递质(如 5-羟色胺/血清素 Serotonin、胃泌素释放肽 GRP、神经降压素 Neurotensin 等)。BON-1 是研究胰腺 NET 肿瘤生物学、抑癌基因(如 MEN1、DAXX/ATRX)功能缺失机制、生长抑素受体(SSTRs)表达与靶向治疗,以及肽受体放射性核素治疗(PRRT)和抗肿瘤药物筛选的国际标准细胞模型。

1. 完全培养基配方 / Medium Formulation
基础培养基 / Basal Medium:DMEM/F-12 (1:1 混合) 基础培养基
血清与添加剂 / Serum & Supplements:10% 胎牛血清 (FBS)
抗生素 / Antibiotics:100 U/mL 青霉素 + 100 ug/mL 链霉素
2. 培养与传代操作 / Incubation & Subculture
培养环境 / Conditions:37 摄氏度,5% CO2,饱和湿度
密度管理 / Confluence:保持细胞密度在 30% 到 85% 之间,避免过密导致细胞脱落或表型改变
传代步骤 / Procedure:
吸除旧培养基,用 PBS 洗涤一次。
加入 0.25% Trypsin-EDTA,在 37 摄氏度下孵育 2 到 4 分钟至细胞脱落。
加入完全培养基终止消化并轻轻吹打。
以 200 x g 离心 5 分钟,重悬后按 1:2 至 1:4 比例接种。

1. Background and Origin
BON-1 (also known as BON) is an immortalized cell line derived from a human pancreatic neuroendocrine tumor (pNET). It retains key biochemical features of functional neuroendocrine cells, including the synthesis and secretion of various bioactive peptides and neurotransmitters (such as serotonin, gastrin-releasing peptide, and neurotensin). BON-1 is a benchmark model for investigating pNET tumorigenesis, somatostatin receptor (SSTR) signaling, peptide receptor radionuclide therapy (PRRT), and targeted drug screening.

Culture Medium Formulation

Incubation and Subculture Guide
Incubation Conditions: 37 degrees Celsius, 5 percent CO2, humidified atmosphere
Confluence Management: Maintain cell density between 30 percent and 85 percent to ensure optimal growth and prevent detachment
Subculture Procedure:
Remove spent media and wash once with PBS.
Add 0.25 percent Trypsin-EDTA and incubate at 37 degrees Celsius for 2 to 4 minutes until cells detach.
Neutralize with complete medium and mix gently.
Centrifuge at 200 x g for 5 minutes, resuspend, and seed at a ratio between 1:2 and 1:4.
