培养与复苏操作规范 / Cultivation & Handling Guidelines
1. 培养基与环境条件
推荐培养基:GC 琼脂基础(GC Agar Base)添加 1% IsoVitaleX 或 Kellogg 营养添加剂;或巧克力琼脂平板(Chocolate Agar)。
培养温度:35 - 37 deg C。
气体环境:3% - 5% CO2 湿润环境(或使用二氧化碳培养箱 / 烛缸)。
生物安全等级:BSL-2(第二类危险微生物)。
2. 复苏与保存规范
冻干株/冻存株复苏:接种于预温的巧克力琼脂平板,划线分离单菌落,在 35-37 deg C、5% CO2 下培养 18-24 小时。淋球菌对干燥和低温极敏感,切勿置于 4 deg C 冰箱过夜保存。
菌种保存:短期保存需每日传代;长期保存建议使用含 15%-20% 甘油的胰蛋白胨大豆肉汤(TSB)或脱脂牛奶,于 -80 deg C 深低温冻存。

General Overview
This reference strain panel represents the official World Health Organization (WHO) 2008/2016 Neisseria gonorrhoeae reference strain set, deposited under the National Collection of Type Cultures (NCTC). Maintained and distributed for standardized quality control, these strains (NCTC 13477, 13820, 13483, 14208, 13818) cover a well-defined spectrum of antimicrobial susceptibility and resistance profiles for AST assay calibration, molecular diagnostics (NAAT/PCR), and Whole Genome Sequencing (WGS) benchmarking.
Technical & Phenotypic Specifications
1. Strain Characterization Matrix
WHO F (NCTC 13477)
Phenotype: Fully susceptible reference control strain.
Key Profile: Low MICs to Penicillin, Ceftriaxone, Cefixime, Azithromycin, Ciprofloxacin, and Spectinomycin.
Application: Baseline reference for MIC determination and culture media QC.
WHO X (NCTC 13820)
Phenotype: Extensively Drug-Resistant Neisseria gonorrhoeae (XDR-NG).
Key Profile: Decreased susceptibility/resistance to Extended-Spectrum Cephalosporins (ESC; Ceftriaxone, Cefixime), fluoroquinolones, and penicillins. Driven by mosaic penA XXXIV allele and mtrR promoter mutations.
Application: QC control for cephalosporin resistance surveillance and ESBL/ESC detection assays.
WHO O (NCTC 13483)
Phenotype: High-Level Azithromycin Resistant strain (HLAzR).
Key Profile: Azithromycin MIC > 256 ug/mL due to homozygous A2059G mutation in all 4 copies of 23S rRNA.
Application: QC standard for macrolide resistance assays and A2059G point-mutation molecular screening.
WHO Q (NCTC 14208)
Phenotype: Multidrug-resistant strain (MDR-NG / PPNG).
Key Profile: Resistant to Penicillin, Tetracycline, and Ciprofloxacin; carries plasmid-mediated beta-lactamase (blaTEM).
Application: QC control for plasmid-mediated penicillinase production and fluoroquinolone target mutations (gyrA/parC).
WHO V (NCTC 13818)
Phenotype: ESC decreased susceptibility reference strain.
Key Profile: Intermediate/elevated MICs for Cefixime and Ceftriaxone via non-mosaic/mosaic penA variants combined with porB1b (penB) efflux/influx alterations.
Application: MIC cut-off validation for oral/injectable cephalosporins in clinical AST workflows.
2. Recommended Culture & Growth Protocols
Growth Medium: GC Agar base supplemented with 1% IsoVitaleX / Kellogg's supplement, or Chocolate Agar.
Incubation Conditions: 35 - 37 deg C under 3% - 5% CO2 in a humidified atmosphere for 18 - 24 hours.
Handling Note: Fastidious organism; highly sensitive to desiccation, temperature drops, and autolysis. Do not store live cultures at 4 deg C.
Biosafety Level: Biosafety Level 2 (BSL-2).
