BioVector® VM7luc4E2 (also known as VM7 or optimized BG1Luc4E2) is a highly sensitive human-derived reporter cell line specifically designed for the detection of Endocrine Disrupting Chemicals (EDCs) and estrogenic activity. Derived from a human breast adenocarcinoma lineage, these cells are stably transfected with a Firefly Luciferase reporter gene plasmid driven by Estrogen Response Elements (ERE). Expressing high levels of endogenous human estrogen receptors ($\alpha$ and $\beta$), VM7luc4E2 is recognized by the US EPA and OECD as a gold-standard in vitro model for assessing the estrogen receptor agonistic or antagonistic activities of chemicals.

2. 培养条件

2. Culture Conditions

  • Maintenance Medium: BioVector® DMEM/F12 (Phenol Red-Free) Medium

  • Serum Supplement: 10% BioVector® Fetal Bovine Serum (FBS)

  • Selection Marker: Maintain with G418 to ensure the stability of the luciferase expression cassette.

  • Assay Preparation: To minimize background signaling, cells are typically starved in medium containing 5% Dextran-Coated Charcoal-stripped FBS (DCC-FBS) for 24-48 hours prior to chemical exposure.

  • Incubation: 37 degrees Celsius, 5% CO2.

3. Applications

4. Key Usage Notes

  • Phenol Red Interference: Phenol red acts as a weak estrogen mimic; therefore, Phenol Red-Free media must be used for all transactivation assays.

  • Hormone Depletion: The use of DCC-FBS is critical during the assay phase to remove endogenous hormones that would otherwise cause high baseline luciferase activity.

BioVector® VM7luc4E2 is extremely sensitive; precautions should be taken to prevent interference from potential leachable endocrine disruptors (e.g., phthalates) found in laboratory plasticware.
