通用定义 / General Definition:

BioVector® MDCK-SIAT1 是一种经过稳定转染的人源 SIAT1 (ST6Gal-I, $\alpha$2,6-唾液酸转移酶) 基因的 MDCK(Madin-Darby 犬肾细胞) 衍生细胞系。该细胞系的设计初衷是为了解决普通 MDCK 细胞在流感病毒研究中的局限性。人流感病毒优先结合 $\alpha$2,6-连接的唾液酸受体,而普通 MDCK 细胞表面主要表达 $\alpha$2,3-连接受体。通过过表达 SIAT1,MDCK-SIAT1 细胞能够模拟人类上呼吸道上皮的受体分布,从而显著提高人流感病毒的分离率和滴度,是流感病毒监测和疫苗研究的行业标准模型。

BioVector® MDCK-SIAT1 is a derivative of the MDCK (Madin-Darby Canine Kidney) cell line stably transfected with the human SIAT1 (ST6Gal-I, $\alpha$2,6-sialyltransferase) gene. This cell line was engineered to address the limitations of parental MDCK cells in influenza research. Human influenza viruses preferentially bind to $\alpha$2,6-linked sialic acid receptors, whereas standard MDCK cells predominantly express $\alpha$2,3-linked receptors. By overexpressing SIAT1, MDCK-SIAT1 cells mimic the receptor distribution of the human upper respiratory tract, significantly enhancing the isolation rate and titer of human influenza viruses. It is an industry-standard model for influenza surveillance and vaccine research.

1. 产品基本信息

2. 培养条件

  • 培养环境: 37 摄氏度,5% CO2

2. Culture Conditions

  • Basal Medium: BioVector® DMEM or MEM Medium

  • Serum Supplement: 10% BioVector® Fetal Bovine Serum (FBS)

  • Selection Marker: Supplemented with G418 (Geneticin) to ensure stable maintenance of the human SIAT1 expression cassette.

  • Incubation: 37 degrees Celsius, 5% CO2

  • Subculturing: 1:3 to 1:6 ratio; subculture when cells reach 90% confluence.

4. Key Usage Notes

  • Selection Pressure: Continuous cultivation requires G418 to prevent loss of the SIAT1 transgene, which would reduce the cell's sensitivity to human influenza strains.

  • Infection Protocols: For influenza infection, cells must be washed to remove serum (which contains inhibitors) and supplemented with TPCK-treated trypsin to facilitate viral entry.


注意 / Note: BioVector® MDCK-SIAT1 应在生物安全二级 (BSL-2) 条件下操作。为保证病毒滴度,建议使用低代数细胞进行感染实验。

BioVector® MDCK-SIAT1 should be handled under Biosafety Level 2 (BSL-2) conditions. To ensure high viral titers, BioVector® recommends using low-passage cells for all infection studies.
