生产原理:

  3. 微环DNA仅含真核表达盒,不含细菌骨架序列,大小仅为亲本质粒的约50%

Production Principle:

  1. Upon L-arabinose induction, ΦC31 integrase mediates recombination between attB and attP sites, generating minicircle DNA and a backbone circle

  2. I-SceI endonuclease cleaves the I-SceI recognition site on the backbone circle, leading to its degradation

  3. Minicircle DNA contains only the eukaryotic expression cassette, devoid of bacterial backbone sequences, approximately 50% the size of the parent plasmid
    Production Advantages:

  4. Simplified procedure, requiring only a temperature change and 5-hour L-arabinose induction

  5. High yield: 3.4-4.8 mg of minicircle DNA per 1000 ml of overnight culture

  6. High purity: contaminating DNA accounts for only 0.4%-1.5%

  7. Low cost: comparable to standard plasmid preparation
    Characteristics:

  8. Bacterial Backbone-Free: Minicircle DNA lacks bacterial replicon and antibiotic resistance genes, avoiding immune responses caused by CpG motifs

  9. High Expression Efficiency: Due to reduced size, transfection efficiency is improved, and transgene expression is more efficient and persistent
