特性特征:

  1. 神经元限制性: 细胞系具有神经元限制性,分化后获得神经元表型

  2. 分化潜能: 在39°C分化条件下,细胞停止分裂,呈现神经元形态,表达神经元特异性烯醇化酶和所有三种神经丝蛋白

  3. 血清素能特性: 分化后表达色氨酸羟化酶;经脑源性神经营养因子处理和部分膜去极化条件(40mM KCl)诱导后,可合成血清素

Derivation: Established by infecting embryonic day 13 (E13) rat medullary raphe cells with a retrovirus encoding the temperature-sensitive SV40 large T antigen (tsA58), followed by infection and clonal selection
Immortalization Mechanism: At the permissive temperature (33°C), SV40 large T antigen is expressed, allowing cell proliferation; at the non-permissive temperature (39°C), SV40 large T antigen is inactivated, cells cease division and initiate differentiation
Morphology: Fibroblast morphology under proliferating conditions; neuronal morphology with neurite extension under differentiating conditions
Growth Characteristics: Adherent; Temperature-dependent growth
Characteristics:

  1. Neuronal Restriction: The cell line is neuronally restricted, acquiring neuronal phenotype upon differentiation

  2. Differentiation Potential: Under differentiating conditions at 39°C, cells cease dividing, exhibit neuronal morphology, and express neuron-specific enolase and all three neurofilament proteins

  3. Serotonergic Properties: Expresses tryptophan hydroxylase upon differentiation; synthesizes serotonin following brain-derived neurotrophic factor treatment and partial membrane depolarization conditions (40mM KCl)

  4. Receptor Expression: Expresses low-affinity nerve growth factor receptor and trkB receptors; expresses high-affinity 5-HT1A receptors with 20-fold increase in Bmax upon differentiation

  5. Transporters: Expresses sodium-dependent 5HT reuptake transporter; expresses synaptic vesicular monoamine transporter

  6. Research Applications: Widely used in serotonergic neuronal differentiation mechanism studies, neurofilament assembly regulation research, neurotransmitter synthesis regulation studies, neurodevelopmental biology research, and cell transplantation therapy studies for chronic pain models
