Characteristics:

  1. Multilineage Differentiation Potential: Can be induced to differentiate into erythroid, myeloid, and lymphoid lineages in vitro -1-4-5

  2. Functional Heterogeneity: The cell population can be separated into subpopulations based on CD34 expression:

    • Sca-1+ c-kit+ Flk- CD34+ cells (approximately 80%) and Sca-1+ c-kit+ Flk- CD34- cells (approximately 20%) -5-7

    • CD34+ cells proliferate in response to SCF and contain the majority of progenitors capable of multilineage differentiation -1-5

    • CD34- cells proliferate in response to IL-3, show poor differentiation capacity, and may represent a more primitive "quiescent" state -1-5

    • The CD34+ population is also heterogeneous: CD34high cells have the highest colony-forming capacity, while CD34low cells retain the ability to revert to a CD34- state -5-7

  3. Self-Renewal: Can be propagated indefinitely in SCF-containing medium and recloned without losing multipotentiality -1

  4. Research Applications: Recognized as an ideal in vitro model for studying early hematopoietic differentiation events, CD34 expression regulation, hematopoietic stem cell activation mechanisms, and gene function in hematopoiesis -1-4-5
