培养指南

细胞形态：上皮样细胞，单层贴壁生长。
培养基：参照 SOP CELL312-SMGM 标准操作程序准备 SMGM 培养基，不添加抗生素。
传代培养：在 T75 培养瓶中接种约 1 x 10的6次方个细胞；每 2-4 天按 1:2 至 1:4 的比例传代。
- 操作细节： 使用 PBS 冲洗两次，随后使用 TrypLE Express 进行消化。
培养条件： 37 摄氏度，5% CO2 (二氧化碳)。
倍增时间：约 2-3 天。
收获密度： T75 培养瓶的最大密度约为 2-3 x 10的6次方个细胞。
冻存条件：使用 Recovery Cell Culture Freezing Medium 进行冻存。

分子遗传学：携带 CDKN2A A148T 杂合突变。经 DSMZ RNA-seq 分析验证，该细胞系还存在 CDH1 和 SMARCB1 基因表达缺失（这是上皮样肉瘤的典型分子特征）。

Cell Culture Data

Morphology: Epithelial-like cells growing in monolayers.
Medium:SMGM medium according to SOP CELL312-SMGM without antibiotics.
Subculture: Seed at approx. 1 x 10 to the 6th cells in a T75 flask; split 1:2 to 1:4 every 2-4 days.
- Procedure: Rinse culture twice with PBS and use TrypLE Express for detachment.
Incubation: 37 degrees Celsius with 5% CO2.
Doubling Time: Approximately 2-3 days.
Harvest Yield: Maximum density of 2-3 x 10 to the 6th cells per T75 flask.
Storage: Frozen with Recovery Cell Culture Freezing Medium.

Scientific Data

Molecular Genetics:CDKN2A A148T heterozygous. Verified by DSMZ RNA-seq analysis, which also confirmed the loss of CDH1 and SMARCB1 gene expression.
Authenticity: STR analysis according to ANSI/ATCC ASN-0002.1-2021 confirms an authentic profile.
Mycoplasma: Negative via PCR assay.
Viruses: Negative for EBV, HBV, HCV, HIV-1, HIV-2, HTLV-1/2, and MLV via PCR.
