The HBPC/ci37 cell line is a conditionally immortalized human fetal brain pericyte cell line used primarily as an in vitro model for studying the human blood-brain barrier (BBB). These cells possess key brain pericyte functions while offering the practical advantage of continuous proliferation in a lab setting. Key Characteristics Organism: Homo sapiens (Human) Tissue Source: Fetal brain pericytes Morphology: Exhibits typical features of brain pericytes Key Trait: Conditionally immortalized, meaning their proliferation can be controlled by temperature manipulation. Immortalization Genes: Carries two specific genes for immortalization: Temperature-sensitive simian virus 40 large tumor antigen (tsSV40T): Promotes proliferation at 33°C. Human telomerase reverse transcriptase catalytic subunit (hTERT): Contributes to indefinite lifespan. Research Use: Used in the development of highly relevant human-based tricellular (with endothelial cells and astrocytes) static transwell or 3D "Brain-on-a-Chip" models of the blood-brain barrier for drug permeability studies. Culture Conditions The conditional nature of this cell line means that culture conditions are critical and must be adjusted based on whether proliferation or differentiation is desired: Maintenance (Proliferation): Temperature: Cells should be cultured at 33°C to maintain the active function of the tsSV40T antigen and promote continuous proliferation. Medium Supplement: Blasticidin S (at a final concentration of 4 µg/mL) must always be added to the culture medium to maintain the expression of the immortalization genes. Differentiation (Functional Assays): Temperature: To induce differentiation and form a functional barrier model, the culture temperature should be raised to 37°C. At this temperature, the tsSV40T antigen becomes unstable and loses its function, turning off immortalization signals. Medium Change: For differentiation, the medium is typically replaced with FBS- and blasticidin S-free differentiation medium.
