The A549/DDP cell line is a cisplatin-resistant subline of the human lung adenocarcinoma A549 cell line. It was developed through continuous exposure to increasing concentrations of cisplatin (DDP) and is a critical model for studying mechanisms of chemotherapy resistance in lung cancer research. Key Characteristics Organism: Homo sapiens (Human) Tissue: Lung (pulmonary adenocarcinoma) Disease: Non-small cell lung cancer (NSCLC) Morphology: Epithelial-like, adherent monolayer cells, often noted to exhibit a more fibroblastic morphology compared to the parental A549 cells. Culture Conditions Culture Medium: RPMI 1640 medium supplemented with 10% Fetal Bovine Serum (FBS) and stable L-glutamine (2.0 mM) is commonly used. Maintenance: To maintain the drug-resistant phenotype, the cells are typically cultured in medium containing a specific concentration of DDP (e.g., 10μM or 1μM, depending on the specific protocol/supplier). This cisplatin is often withdrawn for a few generations immediately before experiments are conducted. Growth Temperature: 37°C. CO₂ Concentration: 5% CO₂ in a humidified incubator. Subculture Routine: Wash cells with PBS lacking calcium and magnesium. Detachment is typically done using Accutase or trypsin/EDTA solution. Subcultivate when 70-80% confluent.
