The KP-N-SI9s cell line is a human neuroblastoma cell line derived from a metastatic lesion in the left axillary lymph node of a patient. It is classified as a substrate-adherent type (S-type) cell line and is widely used in neuroblastoma research to study the different cell phenotypes within tumors. Key Characteristics Organism: Homo sapiens (Human) Disease: Neuroblastoma (originating from the adrenal gland) Tissue Source: Metastatic lesion in the left axillary lymph node Morphology: Substrate-adherent (S-type), with potential to differentiate into smooth muscle phenotypes Growth Properties: Adherent monolayer Doubling Time: Approximately 49 hours Genotype: Contains a heterozygous KRAS p.G12A mutation Immunomarkers: Alpha-smooth muscle actin and desmin-positive Biosafety Level: Typically BSL-1 Culture Conditions Culture Medium: High-glucose DMEM medium, supplemented with 10% Fetal Bovine Serum (FBS). Some protocols use RPMI-1640 with 10% FBS. Growth Temperature: 37°C CO₂ Concentration: 5% CO₂ in a humidified incubator Subculture Routine: Harvest cells using 0.25% trypsin/0.02% EDTA solution when they reach optimal confluence. Handling and Storage Storage: Cryopreserved vials should be stored in the vapor phase of liquid nitrogen or below -130°C for long-term preservation. Thawing: Thaw vials rapidly in a 37°C water bath, then transfer the suspension to fresh complete medium to remove the cryopreservation agent.
