The pSRK plasmid family consists of broad-host-range expression vectors used for tightly regulated gene expression in diverse Gram-negative bacteria. These plasmids are popular because they can function in a wide range of bacterial species and feature a tightly controlled, IPTG-inducible promoter system. Description and Characteristics Vector Type: Broad-host-range expression vector, meaning it is compatible with many different types of Gram-negative bacteria. Backbone Origin: The design is based on the pBBR1 replicon, which is a stable origin of replication. Host Range: Effective in a wide variety of bacteria, including Agrobacterium tumefaciens, Pseudomonas fluorescens, Rhizobium leguminosarum, and Caulobacter crescentus. Expression System: Uses an IPTG-inducible lac promoter system (lacIq-Plac). This system keeps the target gene expression turned off until the inducer chemical, IPTG, is added to the medium. Selection Marker: The specific antibiotic resistance marker varies depending on the variant of the pSRK plasmid. Common Variants pSRKKm: Confers resistance to kanamycin (typically used at 25-50 µg/mL). pSRKGm: Confers resistance to gentamicin. pSRKTc: Confers resistance to tetracycline. Usage and Culture Conditions Host Organisms: Can be propagated in standard E. coli cloning strains like DH5alpha, but is engineered for functional use in other diverse Gram-negative hosts. Culture Medium: Luria–Bertani (LB) medium is standard for E. coli. The specific requirements for different bacterial hosts may vary. Temperature: Typically cultured at 37°C for E. coli, but some specific hosts like Rhodococcus opacus PD630 grow optimally at 30°C.
