The GIST-48 cell line is an adherent human gastrointestinal stromal tumor (GIST) cell line established from a tumor that progressed during imatinib therapy. It is a key model for studying drug resistance due to its specific combination of KIT gene mutations. Key Characteristics Organism: Homo sapiens (Human) Disease: Gastrointestinal Stromal Tumor (GIST), specifically imatinib-resistant. Growth Properties: Adherent monolayer. Morphology: Spindle-shaped/epithelial-like cells. Key Mutations: The line is characterized by two KIT mutations: Primary Mutation: Homozygous KIT exon 11 V560D point mutation. Secondary (Resistance) Mutation: Heterozygous KIT exon 17 D820A missense mutation. Immunomarker: Expresses c-kit (CD117) and CD34. Derived Subline: The GIST48B subline was derived from GIST-48 after drug pressure, resulting in nearly undetectable KIT expression. Culture Conditions Culture Medium: F10 medium supplemented with 15% Fetal Bovine Serum (FBS), serum extender, and bovine pituitary extract. Other protocols use a 50:50 mixture of Dulbecco's Modified Eagle's Medium/Ham's F-12 (DMEM/F-12) with 15% FBS. Growth Temperature: 37°C. CO₂ Concentration: 5% CO₂ in a humidified incubator. Subculture Routine: GIST-48 cells are typically incubated for 120 hours in experiments, suggesting a slow growth rate similar to other GIST lines. They are often split 1:3 when subculturing.
