The NTCC® N2a-APP cell line is a type of neuroblastoma cell line that has been genetically modified to express the human amyloid precursor protein (APP).

  • N2a cells: These are a commonly used cell line in neuroscience research, derived from a mouse neuroblastoma. They exhibit some neuronal characteristics and are relatively easy to culture in the laboratory.

  • APP: Amyloid precursor protein is a transmembrane protein involved in several cellular processes, including cell signaling and adhesion. In Alzheimer's disease, APP is processed abnormally, leading to the production of amyloid-beta (Aβ) peptides, which are thought to play a key role in the development of the disease.

Why is the N2a-APP cell line important?

  • Alzheimer's Disease Research: N2a-APP cells provide a valuable model system for studying the mechanisms of Aβ production and the pathogenesis of Alzheimer's disease. Researchers can use these cells to investigate how different factors affect Aβ production, test potential therapeutic drugs, and study the effects of Aβ on neuronal function.

  • Drug Discovery: N2a-APP cells are widely used in drug screening assays to identify compounds that may inhibit Aβ production or reduce its toxicity.

Key Characteristics:

  • Expresses human APP: The N2a-APP cell line has been genetically engineered to express the human form of APP, making it a more relevant model for studying Alzheimer's disease than using unmodified N2a cells.

  • Produces Aβ peptides: These cells produce Aβ peptides, allowing researchers to study the effects of these peptides on neuronal function and viability.

Limitations:

  • Not a perfect model: While N2a-APP cells provide a useful model for studying Alzheimer's disease, they do not perfectly recapitulate the complex pathophysiology of the disease in humans.

  • Oversimplification: N2a cells are a simplified model of neurons, lacking many of the complex features of neurons in the human brain.
