pORI19 is a plasmid commonly used in Lactococcus lactis, a bacterium often employed in food fermentation. It's a versatile tool for genetic engineering in these bacteria.

Key Features:

  • Origin of replication (Ori): Allows the plasmid to replicate within L. lactis.

  • Erythromycin resistance gene (Emr): Confers resistance to the antibiotic erythromycin, enabling selection of bacteria carrying the plasmid.

  • Multiple cloning site (MCS): Contains various restriction enzyme recognition sites, facilitating the insertion of foreign DNA fragments.

  • lacZα gene: Encodes a portion of the β-galactosidase enzyme, used for blue-white colony screening to identify recombinant clones.

Applications:

  • Gene expression: Introducing foreign genes into L. lactis for heterologous protein production.

  • Metabolic engineering: Modifying the metabolic pathways of L. lactis to improve its performance in food fermentation.

How it Works:

  1. Transformation: pORI19 is introduced into L. lactis cells through electroporation or other transformation methods.

  2. Selection: Cells containing the plasmid are selected by growing them on media containing erythromycin.

  3. Gene cloning: Foreign DNA fragments can be inserted into the MCS using appropriate restriction enzymes and ligases.

  4. Recombinant protein expression: If a gene encoding a protein of interest is inserted into the MCS, the plasmid can drive its expression in L. lactis.
