NTCC® MCF7 AREc32 is a stable human mammary MCF7-derived reporter cell line. It is used to study the activation of the antioxidant response element (ARE), a regulatory element involved in the expression of genes that protect cells from oxidative stress.
Key Features:
ARE-Luciferase Reporter: Contains multiple copies of the ARE sequence linked to a luciferase gene.
Oxidative Stress Response: Activation of the ARE leads to increased luciferase activity, indicating cellular response to oxidative stress.
Drug Screening: Used to identify compounds that can induce or inhibit the ARE pathway.
Mechanistic Studies: Helps to understand the molecular mechanisms underlying oxidative stress response and its role in cancer.
Applications:
Drug Discovery: Screening for potential therapeutic agents that can modulate the ARE pathway.
Toxicology: Assessing the potential of chemicals to induce oxidative stress.
Cancer Research: Investigating the role of oxidative stress in cancer development and progression.
How it Works:
Oxidative Stress: Cells are exposed to an oxidative stressor, such as a chemical or radiation.
ARE Activation: The oxidative stress triggers the activation of the ARE.
Gene Expression: Activated ARE promotes the transcription of genes involved in antioxidant defense.
Luciferase Activity: Increased expression of the luciferase gene leads to higher luciferase activity, which can be measured.
By monitoring luciferase activity, researchers can quantify the level of ARE activation and assess the cellular response to various stimuli.
