P[NF-κB'-NF-κB]1-IFNβmin-driven expression of human TRL1068 heavy and light chains linked via a P2A self-processing peptide and a furin cleavage site (RKRR) with GSG linker (P[NF-κB'-NF-κB]1-IFNβmin-hTRL1068 HC-RKRRP2A-LC-pA). hTRL1068 HC-RKRRP2A-LC was created by annealing light chain and heavy chain fragment of TRL1068 obtained from PCR amplification of TRL1068-LC using OYL172 (5’-TGAAGCAGGCTGGAGACGTGGAGGAGAACCCTGGACCTGAAGCCCCAGCGCAGC-3’) and OYL173 (5’- CTAGTCTAGACTACTACTCGCCCCG-3', XbaI underlined) and PCR amplification of TRL1068-HC using OYL170 (5’-CCCCCCAAGCTTTCGCCCACCATGGACTGGACCTGGAG-3’, HindIII underlined) and OYL171 (5’-AGGTCCAGGGTTCTCCTCCACGTCTCCAGCCTGCTTCAGCAGGCTGAAGTTAGTAGCTCCGCTTCCCCGCCTTTTCCTTTTGCCGGGGCTCAGGG-3’, RKRRP2A sequence underlined), respectively. The resulting hTRL1068 HC-RKRRP2A-LC sequence was then digested and cloned into pYL83 (HindIII/XbaI).
