Origin:
genetically modified cell line established in 1994 from the EBV-positive B-lymphoblastoid cell line ER/EB2-5 (NTCC®-ACC914) by stable transfection with plasmid pmyc-tet, which also encodes the transactivator protein tTA. MYC expression is observed in the absence of tetracycline and is repressed upon its addition.

Risk assessment:
The cell line is positive for EBV (HHV-4) by PCR analysis. Additionally, expression of immediate-early protein BZLF-1 was detected by western blot after stimulating the cells with phorbol ester / sodium butyrate. The infection was classified as potentially lytic associated with a possible production of active viruses. A transmission of EBV during handling of the cells is possible and the cell line is thus categorized biosafety level 2 according to the German Biological Agents Regulation. Furthermore, the cells are genetically modified (EBNA2 expression inducible by estrogen and MYC expression suppressible by tetracyclin), which renders the cells in combination with the EBV infection biosafety level 2..
