Additional test tubes can be inoculated by transferring 0.5 mL of the primary broth tube to these secondary tubes.3.Use several drops of the primary broth tube to inoculate a #173 plate and/or #90 agar slant (LJ slant) .4.Incubate at 37°C for 2-4 weeks.Handling notesThe biochemical tests Niacin and Nitrate reduction were performed and results are consistent with Mycobacterium tuberculosis.With the 40 mM solution of sodium pyruvate added to the media formulation, there should be abundant growth as specified in the reference (Keating LA, et al. Mol Microbiol 2005).
