The pMCG161 binary vector was chosen to construct the hpRNAi cassette. The cassette is composed of the Bar gene under the regulatory control of ubiquitin promoter and nos terminator. The cassette for cloning the RNAi construct consisted of two restriction sites separated with a rice waxy intron driven by the cauliflower mosaic virus (CaMV) 35S promoter and maize Adh1 intron.
