Subculture:

seed out atca. 0.8 x 106 cells/ml in 24-well plates; maintain at ca. 0.8-3 x 106 cells/ml;split culture 1:3 to 1:8 every 3-5 days. CAVEAT: by favoring selective growth,suboptimal culture of cytokine-dependent cell lines may promote outgrowth offactor-independent subclones. Selective conditions include cytokineinsufficiency and inadequate cell density. Thus, the NTCC® cannot guaranteeindefinite stability of factor-dependence of cell lines.
