Origin:

NTCC's HSB-2 established in 1966 from the peripheral blood of an 11-year-old boy with lymphosarcoma that had progressed to acute lymphoblastic leukemia (ALL); cell line was established and initially passaged in newborn hamsters; cells were described as carrying the t(1;7)(p34;q34) involving the LCK and TRB (LCK-TCRB) genes and a submicroscopic del(1)(p32) leading to the STIL-TAL1 (SIL-SCL) fusion gene; one of the oldest immature T cell lines, also known in the literature as CCRF-HSB-2

Morphology:

round to polygonal cells, growing singly or in dense clumps in suspension;

Medium:

90% RPMI 1640 + 10% h.i. FBS

Subculture:

seed out at ca. 0.3-0.4 x 10⁶ cells/ml; maintain at ca. 0.4-1.0 x 10⁶ cells/ml; split saturated culture 1:3 every 2-3 days; maximal density at ca. 1.3 x 10⁶ cells/ml

Incubation:

at 37 °C with 5% CO₂

Doubling time:

ca. 25 hours

Harvest:

cell harvest of ca. 0.8 x 10⁶ cells/ml

Storage:

frozen with 70% medium, 20% FBS, 10% DMSO

Mycoplasma:

initial contamination was eliminated with BM-Cyclin (tiamulin & minocycline), then negative in DAPI, microbiological culture, RNA hybridization and continuously in PCR assays
