Growth instructions Mouse Arg1 reporter constructs cloned into the pGL3 backbone are UNSTABLE when grown at 37 degrees for large-scale preps. We recommend that plasmids be re-transformed and grown overnight at 30 degrees on Amp plates. The next day, scrape the entire plate into 100mL LB-Amp media and grow at 30 degrees (or room temp) for 4-6hr then perform conventional plasmid preps.
