BioVector® WTC-11 (often referred to as WTC) is a highly characterized human induced pluripotent stem cell (iPSC) line. Established by the laboratory of Bruce Conklin at UCSF, it was generated by reprogramming human male skin fibroblasts using non-integrating episomal plasmids. WTC-11 was selected as the foundational line for the Allen Institute for Cell Science, where it serves as the parent for a vast collection of fluorescently tagged organelle reporter lines. Renowned for its stable karyotype, high differentiation efficiency, and exceptional amenability to CRISPR/Cas9 gene editing, WTC-11 is a "gold standard" tool in cell biology, organoid modeling, and systems biology.

2. 培养条件
基础培养基: BioVector® mTeSR Plus 或 Essential 8 (E8)。

培养环境: 37 摄氏度,5% CO2。
传代建议: 建议在汇合度达到 70-80% 时进行传代,使用 EDTA (Versene) 进行团块传代。

2. Culture Conditions
Basal Medium: BioVector® mTeSR Plus or Essential 8 (E8) defined media.
Support Matrix: Feeder-free culture on Matrigel or Laminin-511 (iMatrix-511).
Incubation: 37°C, 5% CO2.
Passaging: Clump passaging using EDTA (Versene) is recommended; typically subcultured every 3-4 days at 1:10 to 1:20 ratios.
